چکیده

Detection and identification of human body fluids represent very important aspects of forensic biology, since in the case of numerous offences, a firm determination of the DNA source (e.g. semen, saliva, epithelium) rather than establishing the DNA profile alone is of great significance. The presently employed routine methods are insufficient: there are no specific tests confirming the presence of saliva, vaginal secretions, menstrual blood or azoospermic semen. mRNA profiling, or identification and detection of body fluids based on the presence of tissue-specific transcripts is a new, promising technique of biological fluid identification. The method is universal, may be specific, and detection of several biological fluids can be simultaneous thanks to the use of multiplex PcR. To date, several dozen mRNA markers characteristic for numerous body fluids have been identified. The present review paper provides a summary of current knowledge on mRNA profiling for the purpose of biological fluid identification.

Identification of forensically relevant body fluids using a panel of differentially expressed microRNAs.

The serology-based methods routinely used in forensic casework for the identification of biological fluids are costly in terms of time and sample and have varying degrees of sensitivity and specificity. Recently, the use of a molecular genetics-based approach using messenger RNA (mRNA) profiling has been proposed to supplant conventional methods for body fluid identification. However, the size ...

RNA Profiling for the Identification of the Tissue Origin of Dried Stains in Forensic Biology.

Examination of crime scene items for biological evidence typically begins with a preliminary screening for the presence of biological fluids in order to identify possible sources of DNA. Conventional biochemical and immunological assays employed for this screening require multiple tests to be performed in a serial manner, can consume a significant amount of valuable evidentiary material, and ca...

Facile semi-automated forensic body fluid identification by multiplex solution hybridization of NanoString® barcode probes to specific mRNA targets.

A DNA profile from the perpetrator does not reveal, per se, the circumstances by which it was transferred. Body fluid identification by mRNA profiling may allow extraction of contextual 'activity level' information from forensic samples. Here we describe the development of a prototype multiplex digital gene expression (DGE) method for forensic body fluid/tissue identification based upon solutio...

Correction: Development of Highly Sensitive and Specific mRNA Multiplex System (XCYR1) for Forensic Human Body Fluids and Tissues Identification

The identification of human body fluids or tissues through mRNA-based profiling is very useful for forensic investigations. Previous studies have shown mRNA biomarkers are effective to identify the origin of biological samples. In this study, we selected 16 tissue specific biomarkers to evaluate their specificities and sensitivities for human body fluids and tissues identification, including po...

Body fluid identification in forensics.

Determination of the type and origin of the body fluids found at a crime scene can give important insights into crime scene reconstruction by supporting a link between sample donors and actual criminal acts. For more than a century, numerous types of body fluid identification methods have been developed, such as chemical tests, immunological tests, protein catalytic activity tests, spectroscopi...
